Definition and template | परिभाषा और template

English

Transcription synthesizes RNA from a DNA template; it does not directly make protein. RNA polymerase reads the template 3′→5′ and builds RNA 5′→3′. The coding DNA strand has the same sequence as the corresponding primary RNA, replacing T with U, when both are written 5′→3′. Different genes may use different DNA strands as templates.

Stages |

English

Initiation involves promoter recognition and opening of a local DNA region. Elongation adds complementary ribonucleotides, and termination releases the RNA. RNA polymerase can initiate without a DNA-replication-style primer. Bacteria commonly use sigma factors for initiation; eukaryotic transcription involves distinct RNA polymerases and transcription factors.

Eukaryotic processing |

English

A typical eukaryotic protein-coding pre-mRNA receives a 5′ cap, undergoes splicing to remove introns, and usually receives a 3′ poly(A) tail. Exons are retained in mature RNA but can include untranslated regions; exon does not mean that every base codes protein. Alternative splicing can produce different transcripts from one gene. Processing details have biological exceptions.

Worked sequence and comparison |

English

Coding DNA 5′-ATG GAA TTT-3′ corresponds to template 3′-TAC CTT AAA-5′ and RNA 5′-AUG GAA UUU-3′. This short example does not include a complete promoter or terminator. Replication copies DNA into DNA, transcription makes RNA, translation reads mRNA to make a polypeptide; distinguish substrate, product and enzyme.

coding DNA 5′-ATG GAA TTT-3′, template 3′-TAC CTT AAA-5′ और RNA 5′-AUG GAA UUU-3′ हैं।

Worked application |

Copying the template sequence unchanged instead of taking its RNA complement gives the wrong transcript.

Self-check & exam revision |

Try answering before opening the answer. /

Does transcription make protein directly? |

No, it makes RNA.

Are all retained exonic bases translated? | सभी exon अनुवादित?

No, exons can contain UTRs.

Exam answer plan: define the term, explain the mechanism, add a labelled diagram or calculation where relevant, state assumptions and limitations.
